Confocal microscopes optical sectioning capability make them the ideal tool
for capturing images for colocalization studies. Colocalization can be used to
measure the percentage overlap between two proteins that have been labelled with
different fluorescent dyes or proteins. Several colocalization software packages
are available and it can be difficult known which one to choose and which calculation method to use. We
favour using Image J or FIJI for our colocalization studies.
There are two basic methods for colocalization analysis which can be found on
the FIJI site.
1.Pixel intensity Spatial Correlation Analysis
2.Object Based Overlap Analysis
